Mouse Subcellular Fractions for In Vitro Assays in Preclinical Drug Development

Animal subcellular fractions, such as microsomes, S9, and cytosol, are used in preclinical drug development as in vitro test systems in assays predictive of in vivo behaviors and conditions. These assays specifically provide information on hepatic and extrahepatic metabolism of a drug compound as part of its ADME (absorption, distribution, metabolism, and excretion) profile. or for species comparison to inform later definitive in vitro or in vivo ADME and drug-drug interaction (DDI) studies.

We offer pooled microsomal, S9, and cytosolic fractions isolated from liver, intestine, kidney, lung, and skin tissue from four different preclinical model strains of mouse (CD-1, BALB/c, C57BL/6, and B6C3F1). Other custom preparations of mouse tissue may be available upon request.

Our pooled mouse liver cell fractions are isolated from hepatic tissue of CD-1, BALB/c, C57BL/6 or B6C3F1 mice. Liver subcellular fractions present a high-quality test system for many in vitro assays. Choose mouse liver microsomes, cytosol, or S9 fraction from male or female pools demonstrating high ezyme activity levels.

We offer pooled mouse intestinal cell fractions isolated from enteric tissue of male CD-1 mouse, using specialized techniques to ensure test systems demonstrating the high enzyme activity levels. Options include matching microsomal, S9, and cytosolic fractions prepared with or without PMSF.

Pooled mouse kidney cell fractions isolated from male CD-1 mouse kidney tissue can be used for in vitro assays exploring renal metabolism. Our options include matching microsomal, S9, and cytosolic fractions, all demonstrating high enzyme activity levels.

We offer mouse lung cell fractions isolated from pooled male CD-1 mouse pulmonary tissue. Options include matching microsomal, S9, and cytosolic fractions demonstrating high enzyme activity levels.

Mouse skin cell fractions are isolated from pooled male CD-1 mouse full-thickness dorsal skin tissue. Options include matching microsomal and S9 fractions to minimize variation, each lot , demonstrating high enzyme activity levels ensuring reliable, high-quality data.

Make sure your microsomes and S9 fractions are properly activated

For best results from assays using microsomes and S9 fractions, we recommend using NADPH regeneration to support high metabolism levels by ensuring that the NADPH levels are not a limiting factor in your incubations. Our preferred system, RapidStartTM, uses an enzymatic reaction that converts NADP to NADPH, which is then used as a cofactor by CYPs and oxidized back to NADP, and the cycle continues ensuring stable NADPH levels throughout the incubation.
