Non-CYP Recombinant Enzymes for Drug Discovery & Development
Recombinant enzymes represent a useful test system for studies requiring high levels of individual enzymatic activities and represent the simplest system available to study enzyme interaction. We are proud to be the exclusive distributor of Cypex Bactosomes in North America. These recombinant enzymes have activity levels similar to those found in vivo and are ideal test systems for preclinical reaction phenotyping and enzyme inhibition studies.
Cypex Bactosomes, recombinant enzymes expressed in E. coli bacteria, come in a standard 1 nmol volume and allow customers to choose specific enzyme activities and various levels of reductase and linearity to best suit their assays. In addition to human and animal cytochrome P450 (CYPs), we also offer other drug-metabolizing enzymes, including SULTs, UGTs, GSTA1 and GSTM1, aldehyde oxidase, aldehyde dehydrogenase, and carboxylesterase 1 and 2.
Other human drug metabolizing enzymes
GSTA1 (1 mg of cytosolic protein)
GSTM1 (1 mg of cytosolic protein)
Aldehyde Oxidase (cytosolic extract expressed in E. Coli) (250 uL)
Human Aldehyde Dehydrogenase (1 mg cytosolic protein)
Human Carboxylesterase 1 (CES1) (1mg protein)
Human Carboxylesterase 2 (CES2) (1 mg protein)
Monoamine Oxidase A (MAOA) (0.5 mL)
Monoamine Oxidase B (MAOB) (0.5 mL)
Recombinant CYPs, UGTs, SULTs, and other enzymes for use in drug discovery and preclinical development assays
Different isoforms of CYPs and UGTs are engineered to enable researchers to distinguish a xenobiotic’s effect on the activity of individual enzymes. Some enzymes, such as CYP3A4 and CYP3A5, are very similar in structure, overlap with regards to target substrates, and are hard to distinguish by LC-MS analysis. Recombinant enzymes, therefore, are most often used to confirm any ambiguity observed in reaction phenotyping studies. In cases of low metabolism, recombinant enzymes can be used at high concentrations to amplify metabolic activity for use in metabolic stability screening and inhibitory potential evaluation.
